The development of computational methods that support human health and environmental risk assessment of engineered nanomaterials (ENMs) has attracted great interest because the application of these methods enables us to fill existing experimental data gaps. However, considering the high degree of complexity and multifunctionality of ENMs, computational methods originally developed for regular chemicals cannot always be applied explicitly in nanotoxicology. This book discusses the current state of the art and future needs in the development of computational modeling techniques for nanotoxicology. It focuses on (i) computational chemistry (quantum mechanics, semi-empirical methods, density functional theory, molecular mechanics, molecular dynamics), (ii) nanochemoinformatic methods (quantitative structure–activity relationship modeling, grouping, read-across), and (iii) nanobioinformatic methods (genomics, transcriptomics, proteomics, metabolomics). It reviews methods of calculating molecular descriptors sufficient to characterize the structure of nanoparticles, specifies recent trends in the validation of computational methods, and discusses ways to cope with the uncertainty of predictions. In addition, it highlights the status quo and further challenges in the application of computational methods in regulation (e.g., REACH, OECD) and in industry for product development and optimization and the future directions for increasing acceptance of computational modeling for nanotoxicology.

Despite the increase in funding for research and the rising numbers of peer-reviewed publications over the past decade that address the environmental, health, and safety aspects of engineered nanomaterials (ENMs), uncertainty about the implications of potential exposures of consumers, workers, and ecosystems to these materials persists. Consumers and workers want to know which of these materials they are exposed to and whether the materials can harm them. Industry is concerned about being able to predict with sufficient certainty whether products that it makes and markets will pose any environmental, health or safety issues and what measures should be taken regarding manufacturing practices and worldwide distribution to minimize any potential risk. Nanotechnology is the creation and utilization of materials, devices, and systems through the control of matter on the nanometer-length scale, that is, at the level of atoms, molecules, and supramolecular structures. The essence of nanotechnology is the ability to work at these levels to generate larger structures with fundamentally new molecular organization. These "nanostructures," made with building blocks understood from first principles, are the smallest human-made objects, and they exhibit novel physical, chemical, and biological properties and phenomena. The aim of nanotechnology is to learn to exploit these properties and efficiently manufacture and employ the structures. Control of matter on the nanoscale already plays an important role in scientific disciplines as diverse as physics, chemistry, materials science, biology, medicine, engineering, and computer simulation. For example, it has been shown that carbon nanotubes are ten times as strong as steel with one sixth of the weight, and that nanoparticles can target and kill cancer cells. Nanoscale systems have the potential to make supersonic transport cost- effective and to increase computer efficiency by millions of times. As understanding develops of the way natural and living systems are governed by molecular behavior at nanometer scale, and as this understanding begins to be felt in science and medicine, researchers seek systematic approaches for nanoscale-based manufacturing of human- made products.
