Everyone conducting analyses using immunological methods should read METHODS OF IMMUNOLOGICAL ANALYSIS. Now available! Three detailed volumes supplying a complete practice-oriented introduction to immunoanalysis. A set in themselves, they provide an extraordinarily comprehensive and up-to-date depiction of these methods. Both specialists and non-specialists will appreciate the broad scope of this set: It covers a wide area of the life sciences, chemistry and medicine, adjusted to the extensive applicability of immunoanalytical techniques. We at VCH feel that there is a definite need for this indispensable new series. Immunoanalysis is widely practised by analysts whose interests or training are not primarily in immunology. They clearly need the guidance that a good reference work provides. Furthermore, the range of immunochemical methods itself is so broad and its techniques so varied that even specialist immunologists and immunochemists find it difficult to assess different possible analytical approaches. They, too, need the support of an extensive reference work. Save 20% by subscribing to the series by 30 June 1993!

Immunological Methods a compendium of basic research techniques being used in one of the largest immunology research institutes, the Basel Institute for Immunology, with particular emphasis given to new methodology. The procedures have been described by individuals judged to be highly expert in their specialties. In many instances the methods developed or adapted to unique uses by the contributors have not previously been described in detail. The book contains 34 chapters covering techniques for detection, isolation, and purification of antibodies (including dansylation, two-dimensional chromatography, isoelectric focusing, polyacrylamide gel electrophoresis, and isotachophoresis); measurement of equilibrium constants (equilibrium dialysis, filtration, and sedimentation); and isotope and fluorescent labeling and detection of cell-surface components. Techniques such as isotope laboratory maintenance; chemical modification of proteins, haptens, and solid supports, and haptenation of viable biological carriers; production of antisera against allotypes and histocompatibility antigens and production of antibody with clonai dominance; histocompatibility and MLR testing; and cell separation by haptenated gels and by velocity sedimentation of rosette-forming cells are also discussed. Other chapters cover detection of antibody-secreting and alloantigen-binding cells; immune responses in vitro and their analysis by limiting dilution; production of T-cell factors; hybridoma production by cell fusion; maintenance of cell lines and cloning in semisolid media; and the mathematical analysis of immunological data.

Immunological Methods, Volume III provides information pertinent to the fundamental aspects of immunological methods. This book presents recombinant DNA technology as applied to immunology. Organized into 25 chapters, this volume begins with an overview of the major histocompatibility complex. This text then examines the use of monoclonal antibodies in the identification of lymphocyte membrane antigens and explains the use of monoclonal antibodies in histocompatibility typing. Other chapters consider the methods in two-dimensional electrophoretic analysis of proteins and another to lymphokines that support B cells. This book discusses as well the production and expansion of cloned B and T cell lines and hybridomas. The final chapter deals with the sophisticated methodology used with particular animal species, namely, birds, sheep, and amphibians to probe specific immunological questions. This book is a valuable resource for immunologists and investigators with extensive experience in using immunological methods. Protein chemists and biologists will also find this book useful.

This two-volume reference details immunological techniques for biologists of all disciplines. Volume I includes a detailed discussion of the tissue culture laboratory. It addresses what the lab needs to be, and the general "housekeeping" procedures involved in tissue culture. Presented next are chapters on specific aspects of tissue culture and hybridoma technology. The book includes a review of bioassays for interleukins, and a series of papers on lymphokines and functional assays in vitro. The section on molecular genetic studies begins with consideration of the choice of strategies for cloning the genes of cell surface molecules. It continues with papers on aspects of molecular biological techniques most closely related to immunology. The final section covers immunochemical techniques. Volume II reviews techniques used with small laboratory animals. It includes papers on specialized procedures with animals. Technical aspects are emphasized through a detailed analysis of effects of ultraviolet light on the immune system. Covered also is antigen detection in cells and tissue. The book addresses the important areas of protein purification using monoclonal antibodies. These two volumes are of great importance to those who use immunological techniques, whether they are immunologists or trained in other disciplines. The book is intended for those in animal science, veterinary science, genetics and cell biology, bacteriology, immunology, pathology, biochemistry, laboratory medicine, and hematology.

Immunological Methods, Volume II compiles procedures that are appropriate for studies in immunology. This book discusses the determination of equilibrium binding parameters of monoclonal antibodies specific for cell surface antigens; two-dimensional gel electrophoresis; and measurements of antibodies specific for DNA. The methods in surface physics for immunology; HLA-DR typing by complement-dependent B lymphocyte lysis; and protein A plaque assay for the detection of immunoglobulin-secreting cells are also elaborated. This text also covers the in vitro production and testing of antigen-induced mediators of helper T-cell function; limiting dilution analysis of precursors of cytotoxic T lymphocytes; and induction of antibody formation in mouse bone marrow. Other topics include the long-term culture and cloning of specific helper T cells; cloning of alloreactive T cells; and enzyme immunoassay for the detection of hybridoma products. This publication is valuable to immunologists and medical practitioners researching on immunological methods.
